Figure 1 illustrates the response detection rates versus the mean background spot counts for both the DFR eq) method (closed circle) and the DFR(2x) method (open circle).
Figure 4b and supplementary figure 4b show the specific response against CMV after subtracting the mean background spot numbers from the mean spot numbers in test wells.
The mean background spot values with and without serum were not significantly different (4.9 and 4.7 per 100,000 PBMC, respectively), suggesting that the presence of serum in the medium does not affect background spot production.
In order to successfully detect these two very low responses the background spot production should have been less or equal to two spots which is below the mean background spot production usually found in an average lab (CIP, unpublished data).
The mean background spot production observed in individual laboratories across all tested donors differed significantly between the participating laboratories and could be very low (0.2 spots per 100,000 PBMCs) to very high (58.1 spots per 100,000 PBMCs).

In fact, in 5,000 simulated data sets, the average response detection rates for at least one of the antigens (k = 2 or 10) were <5% with the DFR eq) method and <1% with the DFR(2x) method across a variety of mean background and experimental spot counts (2 to 50).
For each spot, the mean background value was subtracted from the signal for spots in each subarray.
This means that laboratories with lower background spot counts than the average of the panel will likely have a limit of detection that is lower than 6 sPBMCs100,000 PBMCs.
Telomere length was calculated by the mean telomere spot intensity with mean background subtraction of the respective nucleus on maximum projection images.
The ratio of mean spot signal to mean background variation (signal-to-noise ratio) was clearly lower for Alexa 488 then for the other dyes (namely, 1.25, 1.65, 2.88 and 1.88 for Alexa 488, Alexa 594, Cy3 and Cy5, respectively).
To be scored as positive, a response had to be greater than: a) four times the mean background, b) the mean background plus three standard deviations, c) five spots per well and d) 55 spot-forming cells per million (SFC/M).
