Lines exhibiting the most effective silencing of PiAvr3a were either weakly or non-pathogenic on susceptible potato cv.
However, the most effective silencing occurred at the lower MOI of 0.1 (Onc U6shLacZ: 69.7% silencing; Onc miR-LacZ: 80.0% silencing, P = 0.003).
Two independent shLOXL2 sequences (Supporting Information Table S5a), obtained from SuperArray (Qiagen), were tested in transient transfection assays and shLOXL2-3, giving the most effective silencing (60% reduction of LOXL2 protein level) was used for stable transfections.
At 72 hours post-infection, silencing at the high MOIs was further reduced and the most effective silencing was observed with Onc miR-LacZ at an MOI of 0.01 (80.0% silencing).
At the earliest time point tested (24 hours), the most effective silencing occurred at the highest MOI of 1.0 (Onc U6shLacZ: 52.2% silencing; Onc miR-LacZ: 72.1% silencing, P = 0.0012), whereas the least effective silencing was observed at the lowest MOI of 0.01 (Onc U6shLacZ: 1.3% silencing; Onc miR-LacZ: 11.0% silencing, P = 0.0033).
We found that efficiency of BSMV-induced gene silencing varied greatly with the locations and the stages of inoculation, and inoculation with BSMV on wheat spikes from heading to flowering stage by spike-rubbing five times showed the most effective silencing of PDS in wheat spikes (Table 1).

Tax-interacting-protein 1 siRNA oligo 2 and GEF16 siRNA oligo 2 were found to be the most effective at silencing their respective mRNAs and were subsequently used to evaluate the effects of this procedure on Cdc42 activation.
We selected the shRNA that was the most effective in silencing RANKL (Fig. S4).
Utilization of siRNAs is one of the most effective gene silencing methods and is a promising new approach in the analysis of gene function and gene therapy [ 22- 25].
Among the 3 siRNAs, siRNA derived from the S2 target sequence was the most effective in silencing RANKL expression and was used for further experiments in this study.
The most effective TUCAN-silencing plasmid, pSUPERretro TUCAN3, resulted in an approximately 65% of transcript reduction (Fig. 4A; p = 0.000), and almost complete downregulation at the protein level as shown in Figure 4B.
