Having established that mild digestion with pronase E does not destroy PK-sensitive prion infectivity we sought to optimize conditions for complete digestion of residual PrPC.
Our findings show that mild digestion with pronase E followed by detergent solubilisation and NaPTA precipitation produces a brain fraction devoid of PrPC that contains >70% of prion infectivity.
Native chromatin was obtained by mild digestion with micrococcal nuclease (MNase) of isolated nuclei in 3 mM CaCl2 during different times and the reaction stopped by addition of EDTA (50 mM final concentration).
Mouse islets were dispersed by mild digestion with trypsin-EDTA and the single islet cells were plated on glass cover-slips for [Ca2+]i measurement, or in 96-well cell culture plates for cAMP determination.
First, to overcome the limited loading capacity of genes in human islets, we used mild digestion with accutase.
First, a mild digestion with accutase enzyme was found to be necessary to predistend (ie without dissociation) human islets and enhance accessibility to core cells.

PrPSc is rich in β-sheet structure, insoluble in mild detergents, and partially resistant to digestion with proteinase-K (PK), yielding a N-terminal truncated fragment of 27 30 kDa PrP27-300) [4]–[6].
For fibrous animal tissue (e.g. skeletal muscle), homogenization is often combined with mild proteolytic digestion (e.g. Chance and Williams, 1955).
However, when the data from the ELISA and immunoblot tests using mild protease digestion are compared with that of normal control material it is possible that smaller aggregates of PrP molecules may be present.
