isocratic flow
noun
In liquid chromatography, a mobile phase of constant composition. In contrast to this is the so called "gradient elution", which is a separation where the mobile phase changes its composition during a separation process.

The isocratic flow rate of the mobile phase was maintained at 0.20 mL min−1.
An isocratic flow method with 0.3 mL/min was used; the mobile phase consisted of 98%% (v/v) methanol (0.2 % v/v ammonium formate) and 2%% (v/v) acetic acid (0.2 M, adjusted to pH 6 with ammonia).
Methods: The new isocratic RP-HPLC method (Phenomenex Kinetex 150x4.6mm column and isocratic flow of 80%0.1%% TFA in water and 20% of 0.1% TFA in acetonitrile, 1mL/min) has been developed and validated.
Isocratic flow of the mobile phase, composed of methanol/water/acetonitrile (30 30 40 v/v/v), was employed at a flow rate of 1.0 ml·min-1 with a 10-μl injection volume.
A UPLC method was executed, using a Waters ACQUITY UPLC® HSS T3 1.8 μm, 3.0 × 50 mm analytical column with an eluent of 0.05 M phosphate buffer pH 2.5 with an isocratic flow of 0.8 ml/min.
An isocratic flow method with 0.8 mL/min at 22 °C was used with a mobile phase consisting of 80% of a 0.1%H3PO44 solution and 20% methanol, modified after Werner et al. (Werner et al. 2004).
Radiochemical yields (RCY) were determined by radio-HPLC (Zorbax 300SB-C18, 9.4 × 250 mm 5 μ, solvent: 100 mM tetraethylammonium phosphate pH = 2.24, 1 mL/min isocratic flow) and were cross-checked by two radio-TLC methods (silica, solvent: 0.1 M citrate buffer pH 4 and RP18 on alumina, solvent: 100 mM tetraethylammonium phosphate pH = 2.24 + 20 % ACN).
The proposed method is superior to the previously reported LC-MS methods in terms of the sensitivity and simplicity as the method described herein is based on simple one step protein precipitation for sample preparation and isocratic flow of mobile phase containing acetonitrile:water:formic acid (65 35:0.1%, v/v/v) at a flow rate of 0.2 mL/min only.
