Figure 3 presents the relationship between tissue specificity and proportion of disease-associated genes.
In particular, when exploring the relationship between tissue specificity and disease association, we found this relationship most interesting.
We recognize that the relationship between tissue specificity and SNP frequency could be confounded by our method of detecting SNPs.
The correlation between tissue specificity and disease-association has also been reported by other groups [ 13, 14].
We define a metric called tissue specificity score (TSPS) to quantify the relationship between tissue specificity and TSS nCpG content of genes.
There was a negative correlation between tissue specificity and the M F ratio, showing that narrowly expressed genes had more similar expression levels in the two sexes than ubiquitously expressed genes (P < 0.01 for all tissues except lung and embryo, F-test corrected for confounding effects by expression level; slope estimate β = −0.38 [brain] to −0.99 [liver]).

The lower tissue specificity of H3K4me3-enriched genes is probably a reflection of a general negative correlation between tissue-specificity and gene expression level [ 63, 64].
The above-described connections between tissue specificity, expression level, and expression variance and sex-bias may offer explanations for this rapid rate of evolution.
In order to further characterize the relationship existing between tissue specificity, gene connectivity and disease association, the 15,050 genes were classified as either TS or HK.
We found a positive correlation between tissue specificity of expression and the ratio of non-synonymous to synonymous substitution rate when compared to the chicken orthologue of the gene in question (rs = 0.20, df = 7,342, p < 0.0001, Figure 4).
Previous studies have demonstrated a positive relationship between tissue specificity of expression and evolutionary rate, presumably due to selection against the deleterious pleiotropic effect of mutation restricting the divergence of broadly expressed genes [ 40- 42].
