The fluorescence autocorrelation functions were determined by measurements of about 10 repetitions with 30 sec each.
Dynamic PET (listmode) was acquired for 60 min resulting in 38 frames from 10 to 600 sec duration for the full dynamic dataset and 2 frames each with 600 sec duration 20-300 min and 50-60 min p.i). for dual time point PET (DTP).
Its "best shot" camera feature shoots up to 20 pictures continuously at full-sensor resolution, with each photo 0.1333 sec apart: an average of just 7.5 photos/sec.
To do the subjective test, 3 speech signal samples, each with length 6 Sec, were corrupted with the engaged angle grinder noise under various SNRs.
For these measurements, the parameters selected were: Incubation at 30°C, 50% laser intensity, gain of 86, 2.5 mm laser focus point, 900 sec orbital shaking with 3 mm shaking width, and measurement after each cycle with 0.2 sec measurement time per well, 0.0 sec positioning delay.
All samples were sonicated for 5 min with 10 sec rest after each min. The samples were kept at −20°C until used.
The mixture was denatured in a thermal cycler at 96°C for 75 sec followed by step-down annealing, in 1°C steps with 20 sec incubation at each step, from the optimal annealing temperature (A°) + 9°C to A° + 1°C.
The mixture was then incubated for a period of 80 mins at 37°C and thereafter, the well containing the mixture was washed with washing (phosphate buffered saline (PBS), Tween-20) buffer for a total of 3 times at a volume of 200 μL with 30 sec waiting period each time.
After Hot-start, PCR was done for 33 cycles of 1 min. at 94°C, 1 min. at specified annealing temperature and 2 min. at 72°C with 1 sec extension in each cycle in either a MJ Research mini thermalcycler (Model No. PTC 150) or Perkin Elmer DNA Thermocycler (Model No. 480).
To do the subjective test, 24 speech signal samples each with 6-sec-length were corrupted with the engaged angle grinder noise with various SNRs (0 db to 15 db).
