This combination is well tolerated and is an established regimen in patients with non small cell lung cancer, head and neck cancer, and bladder cancer [24]– [24].
It is well established that regimens enriched in protein decrease hunger sensations and subsequent food consumption in rats and humans [21], [22], [30], [31], [32].
Although this may be explained by patient selection and prognostic factors, it also suggests that that second-line salvage therapy may well have a role in the treatment of recurrent glioblastoma, despite the lack of an accepted or established regimen.
The established regimen for this common ailment is twofold.
For example, HSC numbers expand dramatically following transplantation and it is well established that cytostatic regimens and cytokines can induce their rapid cycling [6], [7].
We hypothesised that the standard streptozocin plus bolus 5FU chemotherapy regimen could be improved in terms of both cytotoxicity and patient tolerance by using an infusional 5FU regimen well established in colorectal cancer management (de Gramont et al, 1997).
The platinum-based chemotherapeutic agent cisplatin (cis-diammine-dichloro-platinum) has been well established in clinical treatment regimens due to its effectiveness on human tumour cells, such as in the context of ovarian, lung, testicular and breast cancer (Kelland, 2007; Lee et al, 2004; Sirohi et al, 2008).
Thus this regimen is well established in the geographical area and this probably explains the continued frequent use of this regimen.
Since the activity of this regimen is well established in man as well as in the murine model, it facilitates interpretation of the results [16].
We therefore decided that, while evaluating these drugs in gastroesophageal cancer patients, we would take the opportunity to explore a different schedule, modelled on the attractive 2-weekly FOLFIRI regimen now well established in colorectal cancer.
In the current study, researchers switched mice who were well established on a long-daily-fast regimen to a weekly pattern of five days on followed by two days off.
