Worms were fed starting in the L2 stage for >48 hr ("strong RNAi"), in the L4 stage for 24 30 hr ("medium RNAi"), or for 12 16 hr for ("weak RNAi") before scoring the viability of embryos laid for a period of 4 hr.
Similarly, using the hPXR DPX-2 agonist assay, we found generally good agreement with the hPXR HepG2 data, although Z,E-fenpyroximate appeared to be a weak hPXR agonist; the cell viability of this compound suggests it is also cytotoxic [median inhibitory concentration (IC50), 0.04; Table 2].
If this condition is not satisfied, no GI50-value exists, i.e., the on-target inhibition is to weak to kill enough cells such that a viability of 50% can be reached.
The weak inhibition of viability after reduction of HSPA8 expression was unexpected considering the log2 ratios from HSPA8-A (-0.885, p = 2.0e-5) and HSPA8-B (-0.446, p = 3.2e-2).
