The only other guest who came close to rivaling his popularity was towering Law & Order actor Fred Thompson, about whom well-wishers jockeyed for position all night (and with whom we photo-opped).

To mask the allocation of treatment, we photo-edited to crop the digital radiograph to include only the fracture site.
In order to better visualize G3Mcs1 motility, we photo-bleached the bud region using a 405-nm laser pulse.
To do so, we used we photo-activated S1P2 morpholino at 24 hpf (see [ Eisenhoffer et al., 2012] for characterization of this method) to knockdown S1P2 after heart development occurred.
First, 30 min post-fusion, we photo-bleached the entire volume of the fused cell (including the ER and the diffusing fluorescent dots [ Figure 5B panel 2]) except the two Golgi, which remained the only source of fluorescence.
Using a localized pulse of 405 nm light, we photo-converted mEos in a rectangular region within the leader bleb a short distance away from the neck and then imaged the redistribution of red fluorescence by diffusion within the bleb and cell body.
