Single-point isosteric mutations of Ser47 and Thr42 situated in the stereospecificity pocket gave rise to variants with doubled enantioselectivity toward pentan-2-ol, in solid/gas reactor.
To test if we can generate a 'super-active' substrate sequence by combining the best substitutions at these positions, we created three variants with double-substitution (FVVLQ↓SGF, TVVLQ↓SGF and VVVLQ↓SGF) and three variants with triple-substitution (FVRLQ↓SGF, TVRLQ↓SGF and VVRLQ↓SGF).
As expected, this helix shift is observed only with shiftable SLI variants but neither with preshifted variants nor with double-stranded variants.
The β-galactosidase expression analysis in B. abortus strains harboring the plasmid constructs showed that the original B. neotomae sodC promoter and its two single-nucleotide mutation variants were significantly less active than the variant with double-nucleotide mutation (Fig. 8).
