A gene library of R. erythropolis NCIMB 11540 in pZErO-2 was screened for oxidases using the plate-based oxidase activity screening method adapted from the group of Turner (Alexeeva et al. 2002).
Following this initial trial, we improved on the manual normalization in four subsequent sequencing experiments by simultaneously purifying and normalizing PCR amplicons using the SequalPrep plate-based normalization kit (Invitrogen), following the manufacturer's instructions.
