Although previous studies using such a vector-based siRNA approach have demonstrated constant gene silencing effects (Lee et al 2002; Miyagishi and Tira 2002; Paul et al 2002), their strategies failed to focus the RNAi effects on a targeted cell or tissue population because of the ubiquitous existence of type III RNA polymerase (Pol-III) activity.
