The commonly used protocol for genome editing in the model strain B. subtilis 168 is the use of the integrative vector (suicide vector) technique, which has a protocol complexity similar to gram-negative genera (Haijema et al. 1996; Rahmer et al. 2015; Wenzel and Altenbuchner 2015).
However, even if the gene is eliminated after transient expression from a viral vector, use of the viral vector is associated with risk of insertional mutagenesis leading to cancer or other cellular dysfunction.
