To illustrate the relationship between FDG uptake and everolimus dose, T/B ratios of single tumours were inverted and the mean T/B ratio of the placebo group was subtracted from these values.
Figure 4 shows the direct relationship between the tumor uptake and the injected dose of 10 in two different xenograft models: group-1 mice transplanted with U87-MG tumor and group-2 mice transplanted with PC3-PIP and PC3-flu tumors.
Results showed that in PTEN null tumour xenograft models, 786-0 and U87-MG, the PI3Kβ inhibitor AZD8186 reduces 18F-FDG uptake at a dose of 50 mg/kg, the same dose which causes tumour inhibition, while it has no impact in a PI3Kα mutant tumour xenograft BT474C.
In agreement with glucose uptake data, low dose of METH (20 µM) increased the expression of GLUT1 protein, whereas 200 µM METH decreased the expression of GLUT1 (Fig 7A F).
