PGE2 concentrations were determined in the control undiluted conditioned media collected at each medium change, which were then pooled for analysis with a commercially available competitive ELISA kit (Cayman Chemical Company, Ann Arbor, MI, USA,) in accordance with the manufacturer's instructions.
Under these conditions, undiluted samples could be directly analyzed with very good recoveries, even for the smallest concentrations tested, as is shown in Table 3. Lateral flow assays are a salient option as point of care tests because they are simple and fast and do not require specialized equipment.
The day 3 conditioned media were also analyzed for SAP as described previously [ 5, 27], with the exception that the ELISA plates were coated overnight at 4°C with mouse antihuman SAP antibody (SAP-5; Sigma, St . Louis MO, USA) diluted 1 1,000 in phosphate-buffered saline (PBS) instead of 50 mM sodium carbonate buffer, and undiluted day 3 conditioned media were assayed.
From each blood sample, 100 µl of undiluted whole blood (per condition) was cultured overnight (18 24 hours) without stimulation (negative control), with phorbol 12-myristate 13-acetate (PMA; 0.1 µg/mL, Sigma)/ionomycin (calcium salt, from Streptomyces conglobatus, 1 µg/mL, Sigma) (positive control) and with each of the 9 TLR agonists (Table S1).
These in vivo observations were taken in to account, and we designed the in vitro experiment with a 3 h time bound period to investigate the metabolic fate of cyanotoxins under anaerobic condition with undiluted whole rumen fluid.
Western blots were performed using whole cell lysates of extracellular N. caninum tachyzoites under reducing conditions probed with undiluted tissue culture supernatants.
