Compared to libraries that contain an unbiased initial sequence, we noted that libraries that contained a single barcode, or a mixture of two barcodes yielded significantly fewer sequences (Fig. 1a and Table 1).
Jorde and Ryman (2007) and Antao et al. (2010) also noted that unbiased estimates of effective size (TempoFs) may come at the cost of precision, with wider confidence intervals than moments estimates.
The implications for patient care and research endeavors are noted and an unbiased assessment of consolidation efforts to date is offered.
