These rhythms are orchestrated by an endogenous circadian clock whose mechanism is based on transcription-translation feedback loops (TTFL) where the translated products of clock genes act to inhibit their own transcription.
Nick translated products, however, hybridized to all probes.
The ORFs with the largest lengths were selected, and the translated products were aligned using ClustalW (Bioedit 7.0).
The translated products of these genes revealed identities to corresponding proteins of Burkholderia vietnamiensis (99 100%%) and Cupriavidus necator H16 (63 89%).
The translated products of transcripts D1A1, D1A2, D2A1, D2A2, D3A1 and D3A2 will retain the genetic messages of the intron which Ds1 inserted in.
As Burkholderia sp. 16 S rDNA gene (accession no. FJ667272.1) exhibited 92% identity to the corresponding gene of C. necator (accession no. CP000090.1), it is not surprising to find that the translated products of Burkholderia sp. PHA biosynthetic genes revealed identities to corresponding proteins of C. necator (63 89%).

S-radiolabeled in vitro-translated products corresponding to full length and the R225* ORFs were obtained with TNT Quick Coupled Transcription/Translation Systems (Promega).
To address this, we carried out trypsin digestion assays on radioactively labelled in vitro-translated products derived from fragments of the BRCA1 cDNA.
The nick-translated products were cleaved at the nicks using T7 exonuclease and S1 nuclease, and end-repaired as described above.
In the proteolytic assays, the in vitro-translated products encoding the A1708V variant showed tryptic digestion profiles similar to those of the wild-type product at all trypsin concentrations tested, whereas the protein products of R1699Q, G1738R, and A1708E were degraded at and above 6 μg/mL of trypsin.
Figure 1D shows that the size of the ATG2-translated proriginatinginating from cDNAs 1 to 4 was identical to that of the faster-migrating product synthesized from cDNAs 1' to 4'.
