This is consistent with the reduction of IGS1-F expression seen in sir2Δ trf4Δ strains compared to sir2Δ, but the effect was variable between fresh transformants and old cells.
GFP was observed most strongly in trichomes and guard cells of transformants and in these cells the fluorescence was localized to the cell wall.
For the culture inoculated with empty vector transformants, the Avicel and cell precipitate seemed compact, whereas the precipitates of the various cultures of Y. lipolytica[chimeric CBHI] transformant appeared swollen and fluffy - possibly indicating changes in the cellulose caused by the transformants (Additional file 1: Figure S2 A).
Equal loading of cellular DNA of the transformant and parental cell lines was indicated by equal sample intensity on the gel following electrophoresis and staining with ethidium bromide.

Similar to yeast (Francisci et al, 2011), galactose-grown transfected mutant cybrids displayed increased viability after 48 h of galactose medium incubation (+2.2 fold relative to mock transformants) and decreased apoptotic cell death after 24 h (up to +2.5) (Fig 2A and B).
