To assess the overall comparability of the pyrosequencing datasets obtained from the GS FLX and Titanium platforms, the average GC content of all reads was determined for each dataset.
While the differences in average GC content can be explained by variations in the sequencing chemistry and protocol used for sequencing on the GS FLX and Titanium platforms, the sharp decline of the GC content present in the longer reads is most probably caused by backfolding.

Paired-end reads were generated using Illumina HiSeq 2000 (n = 6,255,556, 2 kb paired-end library) and 454 GS FLX titanium platforms (n = 1,01,139, 2 kb single-read library) at Beijing Genome Institute, BGI, Shenzhen, Guangdong, China.
