To confirm that Jumu functions as a transcriptional repressor of Ndg in the heart, we overexpressed Jumu protein by crossing UAS- jumu to both TinD-GAL4 and Hand-GAL4 drivers, which caused a dramatic reduction of Ndg reporter expression in the Tin-Lb-CCs (Fig. 5E-E″′; supplementary material Table S3 and Fig. S10) and in the ventral-most Tin-PCs (data not shown).
Staining with appropriate antibodies showed hemisegments with localized increases and decreases in both Svp-CC and Tin-CC numbers, compared with control embryos (supplementary material Fig. S6).
At high temperatures (i.e., above 1,000° C, or 1,800° F) the glass may be conductive enough for booster electrodes of molybdenum, graphite, or tin oxide to be inserted in the tank and provide supplementary heating.
