Plasmid DNA from clones containing gene inserts was prepared using QIAprep spin mini-prep kits (Qiagen, Sussex, UK), according to the manufacturer's instructions, and precipitated, washed thoroughly and resuspended in 10 mmol/l Tris-HCl (pH 8.5).
