Approximately 20% of Anopheles transcripts in dissected samples are absent from whole-body estimates, and only a third of transcripts are recorded across all tissues.

About a third of transcripts, although expressed, do not get translated into proteins [ 11] and the lifetime of proteins can differ by orders of magnitude even for proteins that have similar translation rates [ 9].

As expected from the noisy splicing model we found that a larger proportion of NMD-specific exons (48%) were spliced in minor form (included in less than one third of transcripts transcribed from this gene) compared with non-NMD-single exons (26.1%) (One-sided Kolmogorov-Smirnov test, P = 0.02443).

The regulation model does not require the NMD transcripts to be alternatively spliced transcripts, nor, if they are, need they be the minority form (the transcript isoform that constitutes a small fraction (less than one third) of transcripts from the same gene).

Thus RT PCR confirmation of quantitative microarray data, even if only for a third of transcripts, lends further support to the hypothesis that expression patterns are preserved in archival FFPET, although further work is needed to test this fully, including evaluation of technical replicates in a larger dataset.

Furthermore, differential expression was validated by RT PCR for approximately one-third of transcripts (i.e. fold change expression was similar by both assays).

The third group of transcripts shows unique mixed differential expression patterns under the tested conditions (Fig.  6, columns with blue and red highlighting).
