Data from large animal models have shown that subchondral bone changes such as the upward migration of the subchondral bone plate, intralesional osteophytes, subchondral bone cysts, and a generalized impairment of the osseous microarchitecture below the defect persist for a longer period of time than previously acknowledged (Orth et al. 2013a).
The defects persist during the course of prenatal and postnatal development to demonstrate a constrained dose response relationship with the extent of specific killing of GPT neurons.
Impaired postnatal formation of fissures does not appear to be a result of a developmental delay, because the defects persist throughout later developmental stages.
How long and why this defect persists merits further investigation through follow-up studies.
