For each trial, the test templates were drawn at random from the library (with a uniform distribution) and centered at the arrival times generated by the Poisson process above to form a train of test templates.
The normalization of test templates admits description of each trial with a single SNR, for otherwise SNRs need to be averaged over multiple events.
Note that despite the normalization of the test templates, the detection of EPSCs with variable amplitudes can be effectively simulated by varying SNRs.
With a maximum duration of these test templates being ∼20 ms, this Poisson event generator produced overlapping events with reasonable intervals, as seen in real experimental recordings.
We used the longest template in the dsred series as our control, as it exhibited little termination yet was over a kilobase longer than our longest test templates (Figure 2A, top).
Although synthetic DNA is routinely used in many areas of biomedical research and examples of application of synthetic templates for diagnostic assay validation [12], [23], [24] as well as attempts to create multivalent synthetic test templates [25], [26] can be found in scientific literature, this is the first report of a large scale validation strategy based primarily on synthetic genes.

TTF (test template framework [6]) is an approach that generates unit tests from Z specifications.
From the test template, a striatal volume of 22.3 mm3 was defined.
The detection of EPSC test template was declared correct if the absolute value of the difference between estimated and true arrival times was ≤1.5 ms. Note that this tolerance is significantly smaller than the average duration of test template EPSCs (∼11 ms).
The dual-biotin on the test template was bound to the streptavidin-coated beads (MyOne Beads, Invitrogen, Carlsbad, CA).
For quantification, the test template used was cDNA derived from reverse transcribed RNA isolated 0h, 1h, 3h, 8h, 22h, and 48h after infection.

Since I tried Ludwig back in 2017, I have been constantly using it in both editing and translation. Ever since, I suggest it to my translators at ProSciEditing.
