The ability of Srs2 to unwind an obstructing DNA strand on both the leading and lagging strand template led us to hypothesise that the presence of a single-strand DNA binding protein (such as RPA) or Srs2-interacting partners might influence the unwinding.
This, coupled with the need for higher-throughput marine phage DNA template preparation pipelines, led us to devise and optimize a process, based on a protocol originally designed for coliphage lambda (Promega, Madison WI), for purifying DNA from relatively small volumes (100 200 ml) of crude phage lysates containing approximately 108 phage particles per milliliter (see Methods).
This led us to develop templates for both progress and financial reports that would allow Fellows to include all the necessary details while simplifying the process for them.
For these experiments the cues in the templates led to the recording of significantly more results than in the No Template condition, particularly for the computer-based Study 2, where all of the students recorded results in the templates, but only 15%% recorded them in the No Template condition.
At best, cDNA templates led to shortened 18S sequences.

Bioinformatic search of the Hydra database using mammalian CAD protein sequences as templates lead to the identification of sequence contigs (NW_002159242, NW_002118084, NW_002173794, NW_002105431, NW_002159544) related to vertebrate CAD genes.
