In the current work, development proceeded at first with synthetic DNA template testing to establish proof of concept for the chemistry involved, followed by the design of polymerase chain reaction (PCR -based genomic DNA assays to demonstrate PCR -basedforensic applications.
The four different DNA samples (originating from the Lieshout treatment plant) analyzed for 16S rRNA were also used as a template testing four different primer combinations targeting the pmoA gene.
RFLP tests for each marker and enzyme were conducted according to manufacturer's protocols on 10 μL PCR products from a template test panel consisting of DNA from BW, PO, a BW/PO mix (equal ratio), and a negative using only TE.
