Several activities were organized to achieve these objectives, including inauguration of fixed meetings, a workshop, and developing templates for integration of activities [ 19].
We developed a template to facilitate coding of activities carried out at each HPL team meeting.
Nonetheless, in the interest of rigorous data collection, it would be best if future versions of this recording template could reduce the number of activities for which individual time recording is used.
A previous study [ 18] has shown that in a spatial replicator system with complex formation, a deleterious mutation that destroys both catalytic and template activity of replicators imposes a great disadvantage to parasitic replicators because of the effective dilution of the system by inert molecules.
Indeed, when the template activity of RNA is turned down deliberately (F SFR is changed from 2 to 10 at step 2.4 × 10), which means that an RNA chromosome is further disfavored, the four-gene RNA chromosome no longer rises and the DNA/RNA platform is saved (Fig. 3a).
Thus, ionic environment and DNA dyes could affect DNA denaturation, annealing of oligonucleotide primers to the DNA template and/or activity of Taq DNA polymerase, including its enhanced mutation frequency which could inhibit PCR [ 14].
When programmed with repRNA template, the activity of the solubilized and partially purified tombusvirus replicase was reduced to 23% by QC treatment (Fig. 2C, lanes 3, 8, 9), whereas CPZ had only minor effect (lanes 2, 4, 5) when compared with the DMSO treated sample (lanes 1, 6, 7).
Most interestingly, with the applied partially double-stranded RNA template, the activity of NS5B was substantially delayed.
It is important to be cautious with the interpretation of "non-conventional" RT-PCR products such as chimeric transcripts since their existence can sometimes be attributed to experimental artifacts due to template-switching activities of the reverse transcriptase (RT) or Taq polymerase [ 16].
Results of exogenous RNA template activities using the 3′-ends of plus- and minus-strand RNA indicated that the 5th dpi RdRp could initiate minus-strand RNA synthesis more efficiency than the 11th dpi RdRp.
