Supplementary Figure 3 Spot localization precision and measurement error.
We generated N-step function by raising the voltage from 0 to V in V/(N − 1) millivolt increments, where N is the number of spots and V is the voltage amplitude of the final spot (Supplementary Fig. 1c).
Likewise, the amount of urine (determined by measuring the size of each spot, supplementary Fig. S3) voided by the mutants was greater than that by the controls (129.1±29.9 and 23.9±10.2 µl, respectively; n = 17 or 18, mean±SEM, p<0.01, Student's t-test).
Each probe was spotted 20 times in duplicate giving 1.25, 5 or 20 ng per spot (Supplementary Fig. S1A and B).

Using protein extracts from cells harvested in the exponential phase of growth and 2D-electrophoresis analysis as previously described [17], we detected 40 carbonylated spots, representing roughly 10% of the total number of protein spots (supplementary Fig. S3).
We found that Arp3 co-localized with these actin rich transient spots (supplementary material Fig. S5A,B) and may have been involved in polymerization of actin from these spots.

EHD4, the closest homologue of EHD2, was present in 10% of CAV1-positive spots (Supplementary Figure S2B).
In contrast, we did not find correlations between breakpoints and ribosomal protein genes, small nucleolar RNAs, small nuclear RNAs, and Spo11 hot spots (supplementary table S6, Supplementary Material online), features with no anticipated association with breakpoints.
