Cell counting of five sections each showed that about 40% +/− 16.5% of all osteocytes in the cortical bone matrix of P17 tibiae were β-gal+, the majority of them being located at the endosteal side of the cortical bone shaft (supplementary material Fig. S3A).
Using live-cell TIRF imaging we found that talin-1-GFP localizes to both focal adhesions and MYO10-positive filopodia and in many instances was observed to move together with MYO10 spots along filopodia shafts (Fig. 8b; Supplementary Movies 7).
Other devices, such as springs, flywheels, shafts, and fasteners, perform supplementary functions.
YFP+ bulge progeny were detected in all different hair lineages, including the ORS, layers of the inner root sheath, the hair shaft and the hair germ (Supplementary Figure S4A D).
As in the case of Mg2+-free extracellular solution (Fig. 4C), trains of bpAPs (100 stimuli at 20 Hz) led to hippocalcin-YFP translocation in the dendritic shaft rather than to the spines (supplementary Fig. S1).
Interestingly, a circle of approximately 0.032 mm in diameter was found on the shaft surface of needle S2 (see online supplementary figure S2), which was possibly due to the detachment of a lump in the polishing process during manufacture.
Online supplementary figures S1 and S2 show the SEM images of the shafts of these two types of needle, with a magnification of 400×.
