Universally in the Ferrungulata, Paenungulata, Xenarthra and Ameridelphia where septo-neural transposition takes place, there are supplementary modifications of the lumbar spine that relate to resistance against extension of the spine (Table 6).
Results from this alternative statistical model confirmed the high prevalence of evolutionary motif modification (supplementary material S4, Supplementary Material online).
The conditional knockout (cKO) lines were validated for Snrk genomic locus modification (supplementary material Fig. S7B), and for loss of SNRK protein expression (supplementary material Fig. S7C,D).

These CKO mice exhibit reduced levels of mcm5s2U tRNA modification (Supplementary Fig. 1) and recapitulate many FD hallmarks including a depleted number of pain and temperature-sensing TrkA+ neurons in the DRG, a primary site of Ikbkap expression22,25.
The permeability gradually decreased with the increasing cycles of modification (Supplementary Fig. 25), and reached 3.0 ± 1.2 LMH bar−1 and 64.8 ± 11.3 LMH bar−1 for completely covered PC and PES membranes, respectively (Fig. 5b).
In general, we found little indication of substantial effect modification (Supplementary Table 7).
Downregulated transcripts encode proteins involved in antigen presentation, lymphocyte lineage and signalling, as well as RNA processing/splicing and DNA modification (supplementary figure S1B and table S2).
Forager-upregulated genes showed a more consistent signal for underrepresented terms, for example, GO terms associated with metabolic processes and chromatin modification (Supplementary file 3).
Ten million HPRT + cells electroporated with pJB1-Ase generated five 6TGR colonies, four of which (clones 3A, 3B, 10B1 and 10B3) had AseI fragments indicative of the desired modification (Supplementary Material, Fig. S2C).
LC MS analysis of the intact protein confirmed the successful synthesis and purification of the phospho-cysteine AC variant, and LC MS/MS analysis verified the correct location of the synthesized modification (Supplementary Figures S5 and S6).
