The aim of this study was to develop whole-mount tissue processing methods for 3D in situ visualization of the microvasculature that were also compatible with supplementary labeling for other structures of interest in the tissue microenvironment.
However, by enhancing the training with supplementary labeled data from a related source domain, the model's ability to classify target instances can be improved.
The relative abundance of labelled peptides from the samples was easily detected using the isobaric TMT labels (Supplementary Figure 2, available at Carcinogenesis Online) and overall, 174 differentially expressed proteins were identified upon rWNT5A treatment by TMT-coupled LC tandem mass spectrometry analysis (Supplementary Table 1, available at Carcinogenesis Online).
2D RNA proteome mapping of statistically significant enrichments (two-tailed t-test P value, significance threshold of 0.02 Benjamini Hochberg corrected FDR, from 6 biological replicates of 3 6 litters each) in Gene Ontology (GO) terms and gene groups defined in Supplementary Table 7. Labels are displayed for non-redundant GO cell compartment terms and gene groups.
The development of renal tubules was largely normal judged by calbindin-D28K labeled ureteric bud (UB) and E-cadherin labeling (supplementary material Fig. S3B).
The gt_indel_2 and gt_indel_7 primer sets included an M13 site on the left primer to enable fluorescent labeling in a 3-primer reaction, while the gt_indel_9 left primer was directly fluorescently labeled (Supplementary file 1).
Proliferation was not significantly altered, as assessed by BrdU pulse labeling (Supplementary Fig. S2).
On the other hand, a depletion of histidine residues was typical of all acetylated peptides detected by glucose labeling (Supplementary Figure S8A).
We also simulated local [Ca2+] change using a real immunogold distribution directly taken from the replica labelling (Supplementary Fig. 1a), finding a similar Ca2+ waveform (Supplementary Fig. 2c, d).
Uric acid cement crystals (UACE) then grow on outer surfaces of the stone complex (gray layers in Supplementary Fig. 1c, labeled UACE in Fig. 2c and Supplementary Fig. 4a,b).
After the fragments merge into a stone complex, COM then grows on some outer stone surfaces as cements (COMCE; lime green layers in Supplementary Fig. 1c, labeled COMCE in Fig. 2c and Supplementary Fig. 4a,b), which exhibit crystal-face specific differences in organic matter concentrations (sector zoning) (labeled SZ in Fig. 2c and Supplementary Fig. 4b 26.
