If MCHs carried more than 1 exogenous chr3 copies, would loss of each supernumerary copy provide the cell with growth advantage and permit clonal expansion?
Down syndrome (DS) is a chromosomal aneuploidy characterized by the presence of a supernumerary copy of the whole or part of human chromosome 21 (Hsa21) that gives rise to a syndromic intellectual disability.
Few rDNA copies are sufficient to assure complete disjunction (Appels and Hilliker 1982), and even a single supernumerary copy is sufficient to alter segregation patterns based on rDNA-mediated pairing (Karpen et al. 1988; McKee and Karpen 1990; McKee 1996).
The growth advantage of cells with one supernumerary copy of 3p21 compared to two and with no supernumerary copies compared to one copy was confirmed directly by Bromodeoxyuridine (BrdU) incorporation into the KH39 derived MCHs (figure 6).

Gene rearrangements are commonly explained by a process of "tandem duplication-random gene loss" [ 40], assuming that duplicated portions of the mtDNA can be generated by several processes, and that supernumerary copies of each gene will be selected at random to be subsequently eliminated [ 41].
Mental disability in individuals with Down syndrome (DS) is produced by the increased expression of some of the 231 supernumerary genes on the extra copy of chromosome 21.
