In contrast, we observed a substantially reduced lifespan in mice reconstituted with either Bax −/− Bak −/− (median survival: 190 days, range: 105 357 days, n=20, P<0.0001 versus WT) or Bok −/− Bax −/− Bak −/− (median survival: 195 days, range: 111 402 days, n=22, P<0.0001 versus WT) FLCs.
The new results showed that upon Wnt3 stimulation, β-TrCP-bound p-β-cateninS33/S37/T41 was substantially more in GSK3β KD/GSK3α KD MEFs reconstituted with HA-GSK3βY216F HA-GSK3βY216Freconsthanted winh WT HA-GSK3β or HA-GSK3βY216E, iMEFsing that phosphoreconstitutedus of GSK3βY216 impacts phosphorylated β-cateninS33/S37/T41 recruitment of β-TrCP.
In line with this, the co-IP results showed that upon Wnt3a stimulation, β-TrCP recruitment to β-catenin and ubiquitination of the phosphorylated β-cateninS33/S37/T41 were substantially increased in GSK3β KD/GSK3α KD MEFs reconstituted with HA-GSK3βY216F HA-GSK3βY216Fnstitution whereas16 phosphoreconstitutionic mutant (HA-GSK3βY216E) exerted the opposite effect.
It is also noteworthy that the γ of these PM channels is substantially lower than that of RyR reconstituted into lipid bilayers (184) but similar to that of RyR in the PM of insulinoma cells (γK ∼ 169 pS) (185) (see later).
Furthermore, an analysis of the Lineage marker depleted, Sca+, Kit+ (LSK) cell numbers present in the bone marrow revealed that the proportion of LSK cells (which co-enrich with primitive hematopoietic cells and include long term reconstituting cells) was not substantially changed when Mx-cre THOC5/FMIP flox/flox) mice were treated with poly (I C).
The fluorescence of EGFP was substantially quenched when the apo-form was reconstituted with hemin.
