At high stringency, only wild type TG2 and W241A-TG2 effectively bound to the GTP-agarose; which demonstrates that the W241A mutation has no effect on the guanine nucleotide binding ability of the protein (Fig. 2).
Not only could the differences in definitions and recruitment stringency affect the type of patients recruited, but the timing of recruitment could also contribute.
Stringency for both types of probes was calculated according to [ 64].
It seems appropriate "to tune different levels of regulatory stringency (including regulations on acceptable types of consent) to different types of biological materials according to the interests of the donors involved".
However, at low stringency, in addition to wild type and W241A-TG2; Y516F-TG2 and, to a much lesser extent, C277S-TG2 was found to bind to GTP agarose (Fig. 2).
This type of lowered stringency could be achieved by involvement of multiple cells or molecules in the signaling events, thus the resultant performance of the automata would be much closer to the naturally observable phenomena.
However, the rules above, such as they are, are given in order of increasing stringency: in general search queries of type 1 will return more results than those with 3. The more common an individual's name, the more stringent the rule used must be this is our general observation and recommendation, but does not guarantee improved results.
The ethics review procedure and its stringency should be commensurate with the type of research based on an approximate estimation of potential harm.
Detection following a multiplex PCR is based on solid phase hybridization in microtiter plate format using general and type-specific probes at medium stringency, which makes the detection robust in case of small sequence variations.
As expected, when analyzing a heterozygous sample, the signal for mutation CD8/9+G was similar for both mutant and wild-type probes over the stringency range, with a normalized ratio of ∼0.5 at all stringencies (Fig. 2, A and B).
