This hypothesis is supported by our finding that proliferation of STAT5−/− bone marrow cells plateaued at approximately 500 pg/ml G-CSF, which is the concentration measured in the serum of STAT5−/− mice at steady state (Figure 3B).
If the above scenario is initialised with a random spatial distribution of ants, and with all brood having zero stimulus, the total stimulus will grow at first, before reaching a steady state (Figure 1a).
We also detected slow activated outward currents with maximum activation states at +20 mV for a conductance of 48 nS and an inactivating component that turned on at 80 mV, as demonstrated by the tail current at the steady state (Figure 4c, e).
We observed a significant increase in the number of BrdU-labeled, Lin− sca-1+c-kit+ (LSK) cells both at steady state (Figure 4A) and during hematopoietic recovery (Figure 4B), suggesting that a higher fraction of Id1 deficient LSK cells enter the cell cycle as compared to wild-type cells.
For the experimental analysis of hysteresis, we grew PPK-KO carrying the tetracycline-inducible [23] ppk1 and rel-gfp in medium with increasing concentrations of tetracycline (inducer) and analyzed the distribution of cells expressing GFP by flow cytometry in the stationary phase (steady state) (Figure 6A, going up).
Whereas at 10 min the p-EGFR amount per endosome increased with the endosome area, at 30 min (steady state, Figure 1B), the same mean amount was present in small and large endosomes alike.
For condition I the dynamics from all three alternative models converge to same upper and lower steady states (XT = 95 for upper steady state and XT = 0.0001 for lower steady state Figure 3a. The full model is represented by solid red line (up state) and solid blue line (down state).
We have shown in this paper that the p53-AKT network (the Model shown in Figure 1B), in addition to its potential for bistable behavior, is capable of generating sustained oscillations in p53 and MDM2, and that these oscillations are predicted to occur only within a range of IR intensities and only in the low-p53 (pro-survival) branch of steady states (Figure 2).
It is seen that the model predictions (gray lines) match extremely well both the kinetics (Figure 7A) and the steady-state (Figure 7B) results.
Data for Shaker-expressing cells in Fig. 5 were normalized to the same values as the steady-state figure, and show that |ΔZRF|o in the Cu2+-treated cells was 1.5 times as high as the values shown for the same copper-treated cells in the steady-state.
