The interviews, incidentally, were far more searching than the exams: I had to defend my opinions and answer, on the spot, probing questions on a wide range of subjects, not confined to history.
To address this, we used AFM to image the arrangement of both complementary and non-complementary spotted probe sequences, with or without hybridization to a labeled target sequence, at the single molecule level (Figure 5A).
Thus a high intensity for a G-spot probe cannot be regarded as evidence that its target gene is highly expressed.
Of course the G-spot probe will stand out as an outlier if the other probes in its probeset give a contradictory impression and this will cause no problem.
However, if a G-spot probe has a high value (because on this array G-spot probes happen to have high values) and the other probes in the probe set have high values (because the gene is well expressed) then the G-spot probe will not be excluded in calculations of overall gene expression, even though its value is not affected by the gene, but by the conditions under which the array was treated.
Microarrays were generated by spotting Nexterion HiSens E Sildes (Schott, ordered by Peqlab, Prod. no. 39-1125813, ErlanGermanyrmany) with the miRCURY LNA microRNA array ready-to-spot probe set, 6th gen, human, mouse & rat (Exiqon, prod. no. 208410, Vedbaek, Denmark).
For the FMR1 design chips the correlation for replicated probe spots (probe spots with the exact same probe sequence) is 0.906.
They are omnivores and often can be spotted probing the ground with their long blueish bills, in search of tasty spineless creatures, like worms, to eat.
Spotted probes consisted of cDNA inserts amplified as described in section 2.2, except in a reaction volume of 100 µL containing 1.0 µL eluted phage.
The signal intensities of every three spotted probes were averaged.
