After 3×5-minute washes with blocking solution, specimens were incubated with secondary antibody (Alexa Fluor 488; Invitrogen Carlsbad, CA) for 2 hours at RT. Nuclei were visualized by addition of Hoechst (10 μg/ml, Invitrogen) for 5 minutes after secondary antibody solution.
After being labeled overnight at 4°C with anti-serotonin (diluted 1 10,000 in the above blocking solution), specimens were washed in PBS and reacted for 1 hour (cryostat sections) or 3 hours (whole-mounts) with Cy3-conjugated goat anti-rabbit-IgG.
