Necroptosis plays a role in the modulation of the inflammatory response in the skin and intestine, serves as a backup mechanism to clear pathogens, and is involved in the immunologically silent maintenance of the T cell homeostasis.
Indeed, necroptosis may result in the immunologically silent maintenance of immune homeostasis or, alternatively, may provoke strong inflammatory responses, which may be coupled to the emission of 'danger' signals from necroptotic cells (for an excellent review, see Kaczmarek et al).. Microbial pathogen-associated molecular patterns (PAMPs) activate the immune system.

Another HDAC candidate of interest is Sir2, an HDAC critical for establishment and maintenance of silent chromatin [reviewed in Rusche et al. (2003)].

It should be noted that while the ncRNAS mentioned above are certainly required for the initiation of silencing at their respective targets, it is not yet clear if like Xist, they are dispensable for the maintenance of silent epigenetic state at their target loci.
