It is of note that DC-SIGN expression gradually decreased without expressing Langerin when 3-day-LC-like cells were washed to remove TGF-β1 and further incubated with complete culture medium (CCM) plus GM-CSF (Fig. 3D).
The effect of KCs on LC differentiation was dependent on the differentiation stage since DC-SIGN expression on 5-day-differentiated LC-like cells expressing the highest level of E-cadherin (Fig. 2D) was not decreased when co-cultured with E-cadherin+ KCs (Fig. 3E).
Interestingly, alveolar Mϕ constitutively express the MMR (Wileman et al., 1986), but DC-SIGN expression is only induced in alveolar Mϕ in the lungs of tuberculosis-infected patients (Tailleux et al., 2005), which may influence the disease pattern in an important, but yet to be determined way.

In this experiment, pSUPER-siDC-SIGN expression resulted in a 60% reduction in DC-SIGN mRNA levels compared to the non-specific pSUPER-sip53 control (normalized at 100%).
During the generation of monocyte derived iDC the monocytes were exposed to IL4, which is necessary for DC-SIGN expression [44].
However, the exact mechanism behind the higher MR and DC-SIGN expression is yet to be established, and it is possible that impaired shedding of DC-SIGN and MR at the cell surface could also contribute to this phenomenon.
As iDC mature there is a decrease in their uptake of pathogens and decreased expression of pathogen recognition genes [42], [43] these would explain the reduced DC-SIGN expression and is supported by the presentation of maturation markers at 12 h (Fig. 2).
