She had five stays in psychiatric hospitals in the period from 1936 to 1959 and was subjected to various forms of coercion, such as forced sterilization, cold wet sheet packs and forced injections and was never granted a single talk about the origin or meaning of her psychotic episodes.
Structure space is defined as putative geometric arrangement of β-sheets at the resolution of (i) intrapeptide hydrogen bonds along the fibril axis; (ii) β-sheet/β-sheet packing perpendicular to the axis (e.g. steric-zipper packings, etc).; and (iii) peptide/peptide symmetry describing interpeptide hydrogen bonds.
We show that an inhibitor designed to block the sheet-to-sheet packing in Aβ fibrils is able to cap the heights of these oligomers at ∼4 nm.
The remarkable difference between the structure of the global and local minimum was hydrophobic packing (α-helix/β-sheet packing), which caused an energy barrier between the global and local minimum on the free energy landscape.

The latter are organized into a cross-β core as revealed by the fibre X-ray diffraction pattern that shows a sharp equatorial reflexion at 4.7 Å and a fuzzy reflexion at 10 Å suggesting partial disorder in the β-sheet packing within the fibrils (Figure 3B).
We, therefore, considered if the data presented above can allow discrimination between the different β-sheet packing modes within the fibril architectures described by these alternative classes.
This does not appear to be the case for stacked β-sheet amyloids, possibly because the crystalline nature of the β-sheet packing provides structural stability against deformation caused by dehydration.
They suggest that two key features of the fibril structure are preserved: (1) strand arrangement, which reflects interactions parallel to the long fibril axis, and (2) β-sheet packing, which reflects interactions perpendicular to the fibril axis.
