Luciferase reporter plasmids containing 3' untranslated regions (UTR) of ISCU or random genomic sequence (control) were obtained from SwitchGear Genomics (Menlo Park, CA. A Renilla luciferase expression plasmid (pRL-TK vector, Promega, Southampton, UK) was used as transfection control.
Bim siRNA or a scrambled sequence control was transfected into H1299 cells using the transfection reagent RNAi Lipofectamine according to the manufacture instruction (Invitrogen, Life Technologies, Carlsbad, CA, USA).
Stable lines of DSP-silenced and scrambled sequence controls were selected by means of puromycin antibiotics as described in Materials and Methods.
