Briefly, slides were stained as follows: 15 minutes with primary antibody, 8 minutes with secondary antibodies, 10 minutes with 3,3 diaminobenzine and counterstained with hematoxylin.
The steps included were a 25-minute incubation with Rabbit Polink2+ secondary, a 7.5-minute peroxidase blocking step (3% USP H2O2, with ~0.02% v/v Tween-20 added), a 25-minute incubation with Rabbit Polink2+ HRP conjugated polymer, and a 15-minute incubation with GBI (Cat #: C09-100) 3,3′-diaminobenzidine (DAB) chromogen.
NCS are seen as problematic for alignment and phylogenetic analysis because of their structural constraints, non-randomness of evolution, and mutational changes such as slipped-strand mispairing, stem-loop secondary structure excision/repair, minute inversions, and intramolecular recombination [ 9].
There are also perceived challenges in the use of sequence alignment for phylogenetic analysis of NCS [ 9], as complex mutational processes (slipped-strand mispairing, stem-loop secondary structure excision/repair, minute inversions, intramolecular recombination) are prevalent.
Cells were then incubated with primary antibodies (2 hours room temperature) and secondary antibodies (45 minutes to 1 hour, room temperature) in the same solution, washing with PBS-0.1% Triton X-100 before and after incubation.
For protein measurement, MRK16 anti-Pgp (Kamiya Biomedical) antibody (30 minutes, room temperature) was used followed by 20% normal rabbit serum to block (30 minutes, 4°C) and FITC-conjugated goat anti-mouse secondary antibody (30 minutes, 4°C; Dako).
The solution was poured off and the cells incubated sequentially with anti-COX-2 epitope specific antibody (1 200) (Lab Vision Corporation, Cheshire, UK) or anti-phospho-ERK1/2 (1 100) (Cell Signaling) for 60 minutes, biotinylated secondary antibody (1 200) for 45 minutes, and horseradish peroxidase (HRP -conjugated streptavidin for 20 minutes.
The mean time taken by mothers who had attained primary education was 25 minutes and 46 seconds and for mothers who had attained secondary education 25 minutes and 56 seconds a non-significant difference of only 10 seconds.
Sections were then sequentially incubated with a primary antibody for 90 minutes, a relevant secondary antibody for 60 minutes, and counterstained with DAPI (MP Biomedicals, Santa Ana, CA).
It may be much longer in secondary - 30-45 minutes or even a double lesson if it's an extended brief for English or maths.
