Unfortunately, the reliability of the scores in view of experimental data is rather poor (12).
The HLA-DR staining was scored blindly in view of MSI status of the tumour.
It would perhaps have been useful to use another OHRQoL measure in order to confirm the current study's strong association between xerostomia and OHIP scores – particularly in view of Locker's use of the GOHAI [ 9] and the OHIP in the only previous study to have investigated the relationship – but this was not possible because of time constraints during the data collection.
Only PLAG1 subnetwork overlapped with the height-related OMIM gene subnetwork: 4 of 10 nodes were in the OMIM subnetwork (fold enrichment = 7.8, Fisher's Exact Test p-value = 7.3 × 10−4, EASE score = 0.0063; the zoom-in view of the overlap between PLAG1 subnetwork and the OMIM height gene subnetwork is shown in Figure 2d).
To avoid the scoring bias and in view of a recent paper in which CD133 positivity was quantitatively graded [ 35], we used four-tiered scoring method comprising 0 (totally negative cases), 1+, 2+, and 3+ cases (CD133+ cells covering 1-10%, 11-50%, and 51-100%, of the tumor area, respectively).
A clinically significant response to pilocarpine or placebo will be defined as a ≥2 point improvement in xerostomia NRS score compared to baseline, in view of previous work, where a 20% (2 cm) improvement or more against the baseline NRS score was considered to be a positive improvement [ 21, 22].
That all would choose precisely the same VAS score seems improbable, especially in view of the variety of representational heuristics which individuals are known to employ [ 18].
