Sample pooling is widely used in order to overcome this heterogeneity, however, no objective criteria have yet been defined on how to determine the number of samples to be pooled for representatively profiling a field plot.
Our sample pooling technique collects a composite gas sample from several chambers instead of the conventional practise of analyzing samples from chambers individually, thus reducing numbers of gas samples.
At this stage in the protocol the dye should be silenced; if it is not, unspecific labelling during sample pooling would occur in DIGE.
