In an effort to rule out the possibility of contamination and confirm the pivotal role of hydrogen in lubricity of DLC films, we ran similar tests with the same hydrogen-free DLC in deuterium (an isotope of hydrogen) and achieved essentially the same level of reduction in friction.
By contrast, the levels of LRP and SorLA/LR11 did not show the same level of reduction (Figs. 3A and B).
The same level of reduction was also observed in growth-arrested cells transduced with nonintegrative lentiviral vectors in the presence of Nevirapin, a specific inhibitor of the reverse transcriptase (data not shown).
Noticeably, a recent paper showed that the differential requirement for Vpr is not due to the cell proliferation status, as infection of arrested T-cells by Vpr HIV-1 reduced viral replication by 2-fold compared to Vpr virus [9], which is essentially the same level of reduction observed in proliferating cells.
