If some as yet undiscovered heteroduplex repair system in E. coli could repair 18 nt loops, and if it preferentially repaired either Het I or Het II, reversing the marker configuration would control for this by switching which patch type (e.g. 5') is represented by which Het type (e.g. Het I), when the markers are reversed.
