The reasons for the different responses are unclear, however the rapid decrease in γ-H2AX foci in the RPMI8226 cells in the absence of RAD51 foci suggests that resolution of double strand breaks may not be by homologous recombination repair in these cells.

Recombination could contribute to this mutational effect, because recombination can cause mutations during the resolution of double-strand breaks [ 43].
In the near future, there may be scope to model the effects of novel agents on normal tissue radiation responses through in vitro analysis of the kinetics of DNA repair (e.g., resolution of double-strand breaks measured by γ-H2AX foci or the formation of Rad51 foci) (Rakhorst et al, 2006; Sak and Stuschke, 2010; Redon et al, 2011).
