Another early study by Hooshangi et al., investigating synthetic transcriptional cascades comprising one, two, and three repression stages in E. coli (see Figure 7B), demonstrated that as cascade length increases the overall steady-state response curve steepens (increasing n).
The de-repression stage is characterized by a small increase in expression level of genes encoding cellulases, with similar levels of expression seen in N. crassa cultures transferred to media containing no carbon source or media containing Avicel as the sole carbon source.
Future research can address whether the decrease in transcript abundances at 0.4 mm reflects transient transcriptional repression or stage-specific mRNA degradation.
