A dynamic programming based approach is presented which yields up to three times improvement in efficiency over optimized open loop designs and 35% better efficiency over reported closed loop strategies that focus on optimizing velocities.
Interestingly, potential pseudoknots with decent stability were detected downstream from the gag-pro frameshift site in all four viruses (shown in Table 1); moreover, the detected pseudoknots are all formed by basepairing of a stretch of nucleotides in the loop of the previously reported stem-loop structure with a complimentary sequence 61 83 nt downstream.
Note that in the different crystal CCD structures so far reported, the loop extends beyond the area of the α4-helix.
Alu repeats have been reported within loop attachment regions (LARs, sequences attached to a nucleoskeleton after encapsulation and lysis of cells under physiological conditions) [33]; however, given the prevalence of Alu repeats throughout the genome our SARs are only mildly enriched for Alu repeats (P = 0.0163, 6% enrichment for all Alu sequences; P = 0.0053, 10% enrichment for AluS repeats).

It has also been reported that loops are flexible in nature, which affects the activities of loop-harbouring molecules.
This approach marks a difference from the previously reported closed-loop trials, which considered home-use periods for comparison with in-clinic closed-loop studies (13, 16).
However, despite the manual delivery of a premeal bolus during closed-loop phases, the early postprandial period remains a challenging situation that also was not solved in previously reported closed-loop experiments (14, 15).
