We could not regenerate activity of the BCA + CB[7]· 42a system in this manner because the CB[7]· 42a complex (Ka ≈ 10 M–1) exhibits slow dissociation kinetics (koff ≈ 2 × 10 5 s–1; half-life = 9 h) on the time scale of the experiment.
These fractions also lacked in vivo regenerating activities [ 32].
In tissue homogenates and microsomes this activity was bi-directional, containing both 11β-dehydrogenase (glucocorticoid inactivating) and 11β-reductase (glucocorticoid regenerating) activities, fuelled by NADP(H) as co-substrate, and had a modest affinity (high nM Km) for glucocorticoids (Fig. 1).
The following key parameters for a fast and efficient conversion were found: (1) the NADP(H) concentration, (2) the volumetric activity of 2,5-DKG reductase, (3) the ratio of synthetic enzyme activity to regenerate enzyme activity and (4) the glucono-1,5-lactone concentration.
